چکیده

The effect of D-glucosamine on the metabolism of adenine and uridine nucleotides in Sarcoma 180 ascites tumor cells was investigated as part of an effort to determine the biochemical basis for the cytotoxic effects of glucosamine on tumor cells. Incubation of ascites tumor cells with exogenous glucosamine resulted in a 4-fold increase in the pool size of UDP-N-acetylhexosamine. A large fraction of 14C-labeled uridine was accumulated as radioactive UDP-N-acetylhexosamine when the cells were incubated in the presence of B-glucosamine and labeled uridine. At the same time there was a significant reduction in radioactivity associated with UMP, UDP, UTP, and UDP-glucose, as well as a reduction of the UTP pool to half of its normal level. Glucosamine in the absence or in the presence of low (1 mma) concentrations of glucose caused a significant reduction of the intracellular ATP pool. However, no such decrease in the intracellular ATP level was observed in ascites cells exposed to glucosamine in the presence of 10 mM glucose.

Synergistic Effect of 6-Mercaptopurine and 6-Methylmercaptopurine Ribonucleoside on the Levels of Adenine Nucleotides of Sarcoma 180 Cells1

The synergistic antitumor action of 6-mercaptopurine (6-MP) and 6-methylmercaptopurine ribonucleoside (MMPR) is confirmed with Sarcoma 180 ascites cells. Striking decreases are seen in the adenine nucleotide concentrations in cells incubated for 1 hr with this drug combination, and lesser effects are seen on the guanine nucleotide concentrations. Still greater decreases in adenine nucleotide le...

Relationship between antitumor effect and metabolites of 5-fluorouracil in combination treatment with 5-fluorouracil and guanosine in ascites sarcoma 180 tumor system.

The antitumor activity of 5-fluorouracil (FUra) against ascites Sarcoma 180 was significantly enhanced by coadministration of guanosine, and slightly by adenosine, but not by cytidine or uridine. In advanced ascites Sarcoma 180, guanosine also enhanced the action of FUra, but adenosine, uridine, and cytidine did not. The potentiation of antitumor activity by guanosine was reversed by addition o...

Physiological Disposition and Therapeutic Consequences of Adenine Administered via the Gastrointestinal Tract in Normal and Tumor-bearing Mice.

Each of five ascites-cell neoplasms utilized gastrically administered adenine-8-C14 as a source of purine nucleotides. The disposition of the adenine-8-C14 among selected tissues of normal and tumor-bearing animals was examined. The percentage of the administered isotope that was retained by tumor-bearing animals was greater than that of normal animals, and in the mice with neoplasms the radioa...

Correlations between cytotoxicity, biochemical effects, drug levels, and therapeutic effectiveness of daunomycin and adriamycin on Sarcoma 180 ascites in mice.

The effects of adriamycin and daunomycin on mitotic activity, nucleic acid synthesis, and proliferative activity have been compared in Sarcoma 180 ascites in mice and related to the uptake and retention of the drugs by the ascites cells. The half-life of the drug in the cells after a single injection was 4 hr for daunomycin and 12 hr for adriamycin, while after repeated doses the uptake of the ...
